Brother Agatho began his teaching career at St. Mary's Academy in New Orleans, Louisiana, in 1856 and remained there until 1860 when he was transferred to Christian Brothers College High School in St. Louis, Missouri for four years. He was transferred to St. Joseph, Missouri, in 1868 where he was assigned as the third Director. There he purchased the entire block facing Henry Street between 12th and 13th Streets. He opened a boarding department and upgraded the work of the students so that help wanted advertisements in St. Joseph bore the notation "Brothers' College graduates preferred." Brother Agatho died of pneumonia after only a three day illness. On the day of his funeral, the Cathedral in St. Joseph had "standing room only with Bishop Hogan presiding." He was only thirty-five years old at the time of his death and in his twentieth year as a De La Salle Christian Brother.
